Not really. I wouldn't say the fact that someone is gay makes me uncomfortable because it doesn't. But, feminine men and masculine women kind of bug me.

That all depends. Super flaming people do not make me uncomfortable, but I do find them very annoying. I would not be around a person with mannerisms like that if I have a choice.
